Research profileI measure molecular composition in intact samples and check that the result holds on data the model has not seen. The unit has been a cell or a tissue region. My hands-on measurement experience is multiplexed in situ protein and RNA detection, quantitative fluorescence imaging, single-cell and spatial profiling, and high-dimensional analysis, and I have built each of those into a workflow that runs from tissue and assay preparation through the readout to reproducible analysis code. At PB Immune Therapeutics I built the in-house single-cell, spatial and proteomic analysis capability and ran the therapeutic programs it fed. My current work is deep-learning multimodal models over imaging, transcriptomic and clinical data, including segmentation and model scoring on public iPSC-neuron imaging screens.

Position-Relevant Expertise

  • Multiplexed molecular imaging. GeoMx DSP run end to end, including immunofluorescence staining, fluorescence-guided ROI selection, instrument operation and barcode collection. MACSima cyclic multiplexed immunofluorescence, iterative stain, image and erase rounds on one section. Visium spatial transcriptomics, Olink targeted proteomics, and fluorescence panels up to 24 markers.
  • Quantitative image and high-content analysis. Spinning-disk and point-scanning confocal microscopy, IHC/IF on frozen and FFPE tissue with quantitative image analysis, and segmentation, feature extraction and model scoring on public iPSC-neuron imaging screens, held out at the case level.
  • Mammalian cell and tissue systems. Primary human T-cell isolation, expansion and functional assays; monocyte and dendritic-cell differentiation; mesenchymal stem cell culture; enzymatic tissue digestion, Percoll enrichment, and cell and nucleus isolation across mouse, non-human-primate and human tissue.
  • Scalable and batch-aware workflows. Automated library preparation on the 10x Chromium Connect; donor pooling with SNP demultiplexing so donor and run stay separable; multi-site QC across more than 700 donors; batch integration and held-out evaluation in reproducible Python and R pipelines.
  • Program execution and grant writing. A humanized anti-CD40 program led from candidate ranking to non-human-primate studies and a registered patent in 2024. About $1.3 million across five industry proposals, lead writer on four, always under the institutional PI and not as PI. K99/R00 and LSRF are open to me, and I would apply within the first two years.

Four matched cohorts, 16 donors each, pooled four per lane and demultiplexed by SNP so donor and sequencing run were not confounded. 251,055 cells; custom UMI-aware, BAM-level code resolving PDE4D isoform identity from 5′ reads; consensus NMF programs that repeated across the SCAID atlas of 702 samples; held-out AUC 0.780 for arthritis. Short-isoform overexpression raised the pathogenic fraction and a selective inhibitor lowered it.

Finetuning. LoRA adaptation of an open vision-language model (Gemma 4 12B, mlx-vlm) so that an amyloid pathology tile returns a structured JSON reading, with the schema enforced during decoding and the split held out at the case level. Against the same model zero-shot on 800 held-out tiles, cored-plaque F1 went from 0.12 to 0.44 and cerebral amyloid angiopathy from 0.00 to 0.69. Retrieval. A retrieval-augmented question-answering service written end to end: corpus chunking, BM25 and dense-vector retrieval fused by reciprocal rank, query expansion, and streamed answers that carry their sources.
