Research profileI run binding measurements and use them to decide what advances. On an anti-CD40 antibody program at PB Immune Therapeutics I ran Biacore SPR hands-on for binding kinetics, affinity ranking across a candidate panel, receptor-blocking confirmation, and human against non-human-primate cross-reactivity, with ELISA affinity, competition and epitope assays alongside it. Those numbers selected the lead that went into the primate studies, and the program reached a registered patent in 2024 on which I am a co-inventor. I placed and oversaw the CRO and core-facility work behind it, and I read out what a candidate did in cells by flow cytometry, single-cell profiling and targeted proteomics.

Position-Relevant Expertise

  • Quantitative binding analysis. Hands-on Biacore SPR for association and dissociation behaviour, affinity ranking across a candidate panel, receptor blockade, and human against non-human-primate cross-reactivity. ELISA-based affinity, epitope and competition assays.
  • Protein–protein interaction inhibition. The anti-CD40 program was CD40–CD40L blockade. I also did the functional validation in primary human T cells for a CABIN1-derived core peptide that inhibits calcineurin-mediated T-cell activation.
  • Lead selection and optimization. Quantitative binding and functional measurements used to rank candidates, define a lead, carry it into non-human-primate efficacy studies, and support the therapeutic-antibody patent.
  • CRO and program execution. External studies designed and placed, assay conditions specified to the vendor, incoming data reviewed, milestones tracked, and advancement decided against fixed dates. Non-human-primate studies in three species ran this way.
  • Reading out what a molecule did. High-parameter flow cytometry, single-cell and spatial transcriptomics, and targeted plasma proteomics used to show what a candidate changed in cells and in tissue.
  • Grant writing. About $1.3 million across five industry proposals, lead writer on four, always under the institutional PI and not as PI. K99/R00 and LSRF are open to me, and I would apply within the first two years.

Surface plasmon resonance (Biacore) run hands-on through an antibody lead campaign: association and dissociation rates measured per candidate, affinity ranking across the candidate panel, receptor-blocking confirmed on the same surface, and human against non-human-primate cross-reactivity checked before the primate studies were placed. ELISA-based affinity and epitope analysis, and competition binding to separate candidates that bound the same epitope. Those numbers are what selected the lead.

Finetuning. LoRA adaptation of an open vision-language model (Gemma 4 12B, mlx-vlm) so that an amyloid pathology tile returns a structured JSON reading, with the schema enforced during decoding and the split held out at the case level. Against the same model zero-shot on 800 held-out tiles, cored-plaque F1 went from 0.12 to 0.44 and cerebral amyloid angiopathy from 0.00 to 0.69. Retrieval. A retrieval-augmented question-answering service written end to end: corpus chunking, BM25 and dense-vector retrieval fused by reciprocal rank, query expansion, and streamed answers that carry their sources.
